A lyophilized peptide arrives as a freeze-dried cake at the base of the vial. Reconstitution is the laboratory step that returns it to solution so it can be measured, aliquoted, and recorded. The goal is not speed — it is a solution whose concentration you can state precisely and repeat next week.
Concentration is simply the vial mass divided by the diluent volume you add. A 30 mg vial with 3 mL of diluent gives 10 mg/mL; the same vial with 1.5 mL gives 20 mg/mL. Peptides are long-chain molecules and mechanical agitation can shear them. Let the cake sit for a minute, then roll or swirl the vial between your fingers until the solution is clear. Vortexing and vigorous shaking introduce foam and air, both of which make accurate withdrawal harder.
